Captivate Bio offers a portfolio of GMP-grade small molecules. GMP-grade small molecules are produced under well establish, controlled cGMP guidelines and are used as ancillary reagents in the generation of stem cells and in reprogramming, self-and differentiation workflows for cell therapy applications.

RepSox is a potent and selective inhibitor of the TGF-β type 1 receptor (TGFβRI)/ALK5 that exhibits cell permeability and specifically inhibits ALK5 autophosphorylation with IC50 of 4 nM. When used in conjunction with CHIR99021, LDN193189, and Y27632, among others, RepSox aids in the generation of functional human pancreatic beta cells from human pluripotent stem cells (PSCs) (Pagliuca, et al.).

RepSox has also been shown to contribute to the reprogramming of mouse embryonic fibroblasts to iPSCs (Subramanyam, et al.), in addition to the induction of human fibroblasts into neuronal cells for research on Alzheimer’s disease (Hu, et al). It has also been used as a basal media component for culturing human pluripotent stem cells.

Key Applications:

  • Cellular reprogramming: RepSox can replace the need for the SOX2 gene when inducing pluripotency in adult cells.
  • Adipogenesis: It can induce brown fat formation and "browning" of white fat cells, which may have therapeutic potential for obesity and type 2 diabetes.
  • Cancer research: It has been studied for its ability to induce apoptosis in osteosarcoma cells and suppress tumor growth.
  • Tissue differentiation: It can support the differentiation of retinal pigmented epithelium (RPE) cells from induced pluripotent stem cells (iPSCs).
  • GMP Features
    • Enhanced quality testing and documentation
    • High purity and consistent activity
    • Reduce risk of unwanted genetic material
