Captivate Bio's CET Cocktail pack is comprised of the small molecules used in the CEPT strategy: Chroman 1, Emricasan, and Trans-IRIB (CET). These small molecules have been found to enhance cell survival and promote clonal growth of genetically stable hPSCs.
The CET Cocktail pack is comprised of the small molecule components used in the CEPT strategy: Chroman 1, Emricasan, and Trans-IRIB (CET). The CEPT method is a published combination of targeted small molecules and culture additives proven to enhance stem cell survival and address the key challenges in translational stem cell research. The small molecules in Captivate Bio's CET Cocktail provide comprehensive cytoprotection to overcome cellular stressors inherent in stem cell workflows.
This combination of small molecules has been shown to directly and efficiently enhance pluripotent stem cell survival and promote clonal growth and expansion of genetically stable hPSCs.1 The CEPT method allows for safe, controlled, and efficient strategies for the generation, expansion, differentiation, and banking of stem cell-based therapeutic products. This small molecule combination is also key for optimized embryoid body (EB) and organoid formation, as well as superior cryopreservation of pluripotent and differentiated cells by protecting cells from multiple cellular stress mechanisms in a synergistic fashion.1

The P in the published CEPT supplementation method stands for Polyamine Solution, a standard cell culture additive that supports cell growth. The Captivate Bio Polyamine Solution (Cat. No. CCPM08) is available for purchase separately.
The CET Cocktail is composed of the small molecule components of the published CEPT cocktail (Chen et al. Nat Methods 2021). These 3 small molecules, in combination with polyamine supplement, were shown in the study to improve cytoprotection of a variety of hPSC lines. While the treatment duration may vary across applications, the small molecules in the CET Cocktail are intended to be used according to the CEPT supplementation strategy for improved human pluripotent stem cell survival and viability in culture. (See Chen et al. Nat Methods 2021 for more information on CEPT.)Generally, the duration of each CET (or CEPT) treatment is approximately 24 hours. Dissociated cells are plated in CET-supplemented media, which is replaced with fresh culture media the following day during the first feeding. Some clonal assays may require up to 3 days of treatment in order not to disturb the clonal colonies with an excessive media change during the initial attachment period.
