The CET Cocktail pack is comprised of the small molecule components used in the CEPT strategy: Chroman 1, Emricasan, and Trans-IRIB (CET). The CEPT method is a published combination of targeted small molecules and culture additives proven to enhance stem cell survival and address the key challenges in translational stem cell research. The small molecules in Captivate Bio's CET Cocktail provide comprehensive cytoprotection to overcome cellular stressors inherent in stem cell workflows.

This combination of small molecules has been shown to directly and efficiently enhance pluripotent stem cell survival and promote clonal growth and expansion of genetically stable hPSCs.1 The CEPT method allows for safe, controlled, and efficient strategies for the generation, expansion, differentiation, and banking of stem cell-based therapeutic products. This small molecule combination is also key for optimized embryoid body (EB) and organoid formation, as well as superior cryopreservation of pluripotent and differentiated cells by protecting cells from multiple cellular stress mechanisms in a synergistic fashion.1

Alternative names: CE(P)T cocktail, CEPT, cet cocktail molecules
Applications: Stem cell survival, CEPT Cocktail method, cell translation
Cell types: Pluripotent Stem Cells, iPSCs, ESCs, neurons

Features & Benefits

  • Small molecule components of published CEPT cocktail specifically selected for enhanced cell survival.
  • Provides comprehensive cytoprotection to overcome cellular stressors inherent in stem cell workflows.
  • Demonstrates remarkable effectiveness to improve stem cell viability and cloning efficiency after single-cell dissociation.1

The CET Cocktail pack includes the following powder components:

  • Chroman 1 (5 mg) - a highly potent and selective ROCK inhibitor
  • Emricasan (10 mg) - a potent and irreversible pan-caspase inhibitor
  • Trans-ISRIB (10 mg) - an integrated stress response (ISR) inhibitor

For best results, prepare the complete CEPT cocktail according to the published recipe, include Polyamine Solution in addition to the CET Cocktail pack of small molecules in the cell culture media just before use.

CET Cocktail hPSC survival data CEPT Cocktail clonal growth of hPSCs

Figure 1. Cell survival. hPSCs were seeded as single cells at an ultra-low density of just 25 cells/cm2. Data represents various Y27632, Chroman, Emricassan, and Trans-ISIRB small molecules with a polyamine solution. Improvements are seen in cell survival when using CET and CEPT combinations over Y27632. Chen, Y. et. al. 2021.

Figure 2. Improved clonal growth and expansion of hPSCs. An alkaline phosphatase staining assay of hPSCs cultured with Y27632, the complete CEPT combination, and commercially available CloneR™ supplement, clearly showed more alkaline phosphatase-positive clones in cultures exposed to CEPT.

FAQs

What is the P in CEPT?

The P in the published CEPT supplementation method stands for Polyamine Solution, a standard cell culture additive that supports cell growth. The Captivate Bio Polyamine Solution (Cat. No. CCPM08) is available for purchase separately.

Which cell lines has the CET Cocktail been validated with?

The CET Cocktail is composed of the small molecule components of the published CEPT cocktail (Chen et al. Nat Methods 2021). These 3 small molecules, in combination with polyamine supplement, were shown in the study to improve cytoprotection of a variety of hPSC lines. The following cell lines were used in the published study:

  • hESC lines: WA01 (H1), WA07 (H7), WA 09 (H9), WA17, WA14 (H14), WA26, HUES53
  • hiPSC lines: LiPSC-GR1.1, NCRM5, ESI-035, GM25256, GM23476, GM2610, JHU078

How long should cells be treated with the CET cocktail?

While the treatment duration may vary across applications, the small molecules in the CET Cocktail are intended to be used according to the CEPT supplementation strategy for improved human pluripotent stem cell survival and viability in culture. (See Chen et al. Nat Methods 2021 for more information on CEPT.)Generally, the duration of each CET (or CEPT) treatment is approximately 24 hours. Dissociated cells are plated in CET-supplemented media, which is replaced with fresh culture media the following day during the first feeding. Some clonal assays may require up to 3 days of treatment in order not to disturb the clonal colonies with an excessive media change during the initial attachment period.

Can I purchase the CET small molecules separately from the CET Cocktail kit?

Yes. Each of the small molecule compounds in the CET Cocktail can be purchased separately: Chroman 1 (SML20A); Emricasan (SML21B); Trans-ISRIB (SML22B).

What are the components of the CET Cocktail kit?

The CET Cocktail kit contains one vial each of the following small molecules: Chroman 1 (5 mg); Emricasan (10 mg); Trans-ISRIB (10 mg).
