Y-27632 dihydrochloride is a highly specific inhibitor of Rho-associated coiled-coil protein kinase (ROCK) activity as a cell-permeable, ATP-competitive inhibitor of both ROCK1 (Ki of 220 nM) and ROCK2 (Ki 300 nM) (Ishizaki, et al.). Human embryonic stem cell (ESC) survival is improved through enhanced attachment following single cell dissociation, cryopreservation, and FACS sorting in both feeder-dependent and -independent cultures (Emre, et al.). This enhanced cell attachment, and thus recovery, is boosted through decreased dissociation-induced apoptosis (Watanabe, et al.). Y-27632 dihydrochloride primes human induced pluripotent stem cells (iPSCs) to selectively differentiate towards mesendodermal lineage via epithelial-mesenchymal transition-like modulation.

Y27632 is a small molecule inhibitor of the Rho-associated coiled-coil containing kinase (ROCK) that improves survival of human pluripotent stem cells. Y27632 improves cell survival by blocking the cell contraction that leads to cell death by apoptosis and anoikis by inhibiting the ROCK pathway.
